I want to put 5 buttons in scene one

do I need to create a new layer for each button.

E.G LAYER 1 BUTTON 1
LAYER 2 BUTTON 2

Not unless you are going to be placing them one on top of the other.

One layer can contain as many symbols or objects as you can fit onto it, the layers just help for allowing access to certain things on top of others.
